Evaluation of Sperm Chromatin/DNA Integrity, Morphology and Catsper Expression on Diabetic C۵۷BL/۶ mice

چکیده مقاله:

Background: Diabetes is associated with reproductive impairmenton the male reproductive system and causes complicationssuch as decreased libido, fertility, spermatogenesis, spermmotility, and morphology. High level of blood sugar may affectsperm quality and reduces the potential for male fertility. Increasedlevels of sperm DNA damage is often associated withreduced count and motility or abnormal morphology.Materials and Methods: This experimental study was performedin Mashhad University of Medical Sciences. ۴۰ mice(C۵۷BL/۶) were divided randomly into ۴ groups: ۱. Controlgroup, ۲. Diabetic group, ۳. Diabetic + Insulin group, and۴.Sham group. After ۳۵ days, the right epididymis of all specimenswas used for Real-Time PCR and left epididymis forevaluation sperm parameter using Aniline blue, Toluidine blue,Papanicolaou, and immunohistochemical study. Also, right testeswere applied for immunohistochemical and tunnel study.Results: Results of this study showed that chromatin integrity,morphology, and cation channels of sperm (Catsper) expressionwas significantly changed in diabetic mice in comparisonto other groups (P<۰.۰۵) and treatment with insulin improvedthese parameters.Conclusion: Our findings showed that the sperm parameterssuch as DNA integrity, morphology, and Catsper expressionchange in diabetic mice.
