Anti-Apoptotic Effect of Royal Jelly Against Nicotine Induced Testicular Injuries in Mice: Evidence of P۵۳, Bcl-۲ and Caspase-۳ Expressions

چکیده مقاله:

Background: Nicotine (NIC) as a toxic component adverselyaffects a male reproductive system. This study describes theeffects of royal jelly (RJ) on testicular injury induced by NICand elucidates the potential underlying mechanism at molecularlevel.Materials and Methods: Thirty-six male BALB/c mice wererandomly divided into six groups (n=۶). Group ۱ received normalsaline, group ۲ received ۱۰۰ mg/kgBW/day RJ, groups ۳and ۴ received NIC at doses of ۰.۵۰ and ۱.۰۰ mg/kgBW/day, respectivelyand groups ۵ and ۶ received NIC at doses of ۰.۵۰ and۱.۰۰ mg/kg BW/day respectively plus RJ. Following ۳۵ days,the expressions of P۵۳, Bcl-۲ and caspase-۳ in testicular tissuewere evaluated by RT-PCR.Results: NIC treatment caused a significant (P<۰.۰۵) downregulationof Bcl-۲ expression and up-regulation of P۵۳ andcaspase-۳ expressions versus control group. Nevertheless, RJco-administration led to significant up-regulation of Bcl-۲ expressionand down-regulation of the expressions of P۵۳ andcaspase-۳ versus NIC-exposed mice.Conclusion: Our results confirmed that RJ effectively protectstestis against NIC damages through its anti-apoptotic effectsand mitochondria-dependent apoptotic pathway prevention.
