Metformin Attenuates Cognitive Impairment and Anxiety in Ovariectomized Mice

Background: Estrogen regulates many processes in the brainsuch as synaptic formation, learning, and memory. Empiricalevidence shows that there is a correlation among menopause,memory impairment, and anxiety due to Estrogen deficiency.Regarding the neuroprotective and anti-inflammatory propertyof Metformin (Met), the present study was designed to evaluatethe effects of Met on tactile learning and anxiety in ovariectomized(OVX) mice.Materials and Methods: For this purpose, Met (۷ and ۱۵ mg/kg/p.o.), was administrated daily in OVX mice for ۲۱ days. Tactilelearning and anxiety like-behavior were evaluated by novelobject recognition task and Elevated Plus-maze, respectively.Results: Our findings showed that Met prevented from the deleteriouseffects of ovariectomy on learning memory (۷ and ۱۵mg/kg). We also found the improvement in anxiety by increasesingOpen Arm Time (OAT%) and Open Arm Entries (OAE%)in OVX animals treated with Met at the both doses compred tothe OVX group.Conclusion: Collectively, the results of present study suggestthat Met is an convenient choice in menopause for ameliorationof memory dysfunction.
