In Vitro Maturation and Fertilization in Mice NMRI: Comparison of The Effect of Bovine Serum Albumin, Fetal Bovine Serum, Autologous and Allogeneic Mice Serums

Background: Oocyte maturation is a physiological event thatis essential for the successful fertilization and development ofthe embryo. Sera are a complex combination of factors such asgrowth factors, proteins, vitamins, small elements, hormones,etc. which are essential for the growth and maintenance of cellsand can affect oocyte maturation and fertilization. The aim ofthis study was to evaluate the effect of different sera on in vitromaturation and fertilization in NMRI mice.Materials and Methods: Maturation of oocytes was investigatedin Alpha MEM medium with ۲۰, ۱۵, ۱۰, ۵, ۰ allogenicand autologous mice serums, fetal bovine serum (FBS) andbovine serum albumin (BSA) after ۱۴ to ۱۸ hours incubation.Then mature oocytes were fertilized in different sera with a capacitysperm in MHRM medium with percentages of ۲۰, ۱۵, ۱۰,۵, ۰ in different sera.Results: The results showed that the level of in vitro maturationincreased significantly by adding serum to the culture medium(P<۰.۰۰۱). Also, the rate of in vitro fertilization increased significantlyby adding serum to the culture medium (P<۰.۰۰۱).Among all groups, FBS ۱۰% (۸۰%), BSA ۵% (۷۵%), Allogeneicmice serum ۲۰% (۵۴%), and autologous mice serum ۲۰%(۴۰%); were found to have the highest maturation; BSA ۵%(۷۷%), FBS ۲۰% (۷۵%), in Allogeneic mice serum ۲۰% (۷۰%)and in autologous mouse serum ۱۵% (۶۴%), the highest fertilizationrate was observed.Conclusion: There are limited studies on the effects of differenttypes of serum added to the culture medium on different stagesof maturation, fertilization, and subsequent development of theoocyte. However, using inhuman serum can be a source of contaminantsand infections. The results of this study showed thatalthough BSA and FBS had better results than allogeneic andautologous sera, but it was better to use my own serum due tothe possibility of transmission of infections and diseases fromanimals to humans and the need to reduce the costs of infertilitytreatment, it is better to use one’s own body or the same species.
