چکیده مقاله:
Background: The histone modifications are epigenetic marksthat control the developmental pattern of the preimplantationembryo and are influenced by many factors, such as the spermand oocyte quality and gamete in vitro manipulation. The purposeof this study was to investigated the changes in H۳K۴me۳(Histone ۳ lysine ۴ trimethylation )and H۳K۲۷me۳ ( Histone ۳lysine ۲۷ trimethylation ) marks after long term in vitro spermincubation at ۳۷°C.Materials and Methods: Mouse sperm samples incubatedat ۳۷°C for ۲۴ hours. Before and after extended incubationH۳K۴me۳ and H۳K۲۷me۳ marks were evaluated by flowcytometry.Relative expression levels of histone methyl-transferaseH۳K۲۷ (Ezh۲) and histone methyl-transferase H۳K۴(Mll) were assessed by quantitative real-time polymerase chainreaction (qRT-PCR).Results: H۳K۴me۳ and H۳K۲۷me۳ marks were significantlyhigher after incubation at ۳۷°C compared with fresh sample(P<۰.۰۵). The level of two histone methyl-transferases genes(Ezh۲ and Mll) showed significant increase between two groups (P<۰.۰۵).Conclusion: The result of this study demonstrated that extendedsperm incubation affect correct histone methylation patterns.Aberrations in these epigenetic marks may have detrimentalconsequences for early embryonic development.
