Effect of Human Seminal Plasma on In Vitro Growth of Mouse Follicles and Oocyte Maturation

Background: In vitro growth and maturation of pre-antral ovarianfollicles is as one of the important techniques in ART .Thisis a good way to preserve fertility in women whom reducedtheir ovarian function due to various reasons such as radiotherapyand chemotherapy.The current challenges for follicle cultureare optimization of culture media to match the physiologicalneeds of the cell in vivo. The present study was designed todetermine whether different seminal plasma concentrations inthe culture medium could improve the developmental growthof follicles and oocyte maturation. Materials and Methods: Human seminal plasma was obtainedafter centrifugation (۳۰۰۰ g⁄min for ۱۰ min). The culture mediumwas consisted of α-MEM supplemented with ۱۰% FBS,۱۰۰ mIU/ml (rFSH),۰.۰۵,۰.۱ and ۰.۵% (v/v)concentration ofseminal plasma(treatment I ،II وIII respectively)andcontrolgroup, non-seminal plasma.All selected Pre-antral follicles(۱۰۰-۱۴۰μm) from different treatment group were mechanicallyisolated from the ovaries and cultured in different dishesunder the same culture conditions (۵% CO۲ in air at ۳۷°C for ۹days).The follicles from the four experimental groups were assessedfor viability with trypan blue staining, follicles growth,oocyte survival and oocyte maturation rate in vitro. Chi- squaretest was used for evaluating differences between control andexperimental groups.Results: Result indicated that follicle growth rate in differentconcentration of seminal plasma decreased significantly comparedwith control group over a period of ۹ days.Percentage ofoocyte maturation in ovulated oocytes from cultured follicleswith ۰.۱ percentage concentration of seminal plasma increasedsignificantly in comparison of control group(P≤ ۰.۰۵).Conclusion: Seminal plasma is a dose- dependent manner thathas a positive effect on in vitro maturation of oocytes.
