چکیده مقاله:
Background: The paucity of information on seminal characteristicsand their alteration at different holding temperature andholding time led to the present study to provide more data requiredfor efficient in vitro storage of partridge semen.Materials and Methods: Sixty male Chukar partridges werehabituated to abdominal massage for semen collection. Theejaculates from males were pooled and extended (۱ to ۵) inSexton’s diluent. The samples were kept at ۴-۵ºC or ۱۹-۲۴ºCholding temperature for ۴, ۲۴, or ۴۸ hours. The percentage ofmotile sperm, live sperm (eosin-nigrosin staining), and concentrationof thiobarbituric acid reactive species (TBARS) weredetermined in triplicate. The data were analyzed by the mixedprocedure of STATA software.Results: Data showed that the percentage of sperm motility andsperm viability was higher (۸۱.۷ and ۹۱.۶%, respectively) atlower holding temperature and shorter duration of storage time.Also, a lower value of seminal TBARS was recorded at the lowholding temperature and shorter storage time.Conclusion: The percentage of sperm motility and viability washigher and the concentration of TBARS was lower at the lowesttemperature and storage time applied in the current study. Furtherimprovement in the quality of the extended Chukar semenis needed before in vitro preservation over ong periods can becan be recommended for artificial insemination.
