Background: Male sterilization by chemical agents is a nonsurgicalcontraceptive approach designed to induce azoospermiaand, therefore, infertility by light and transmission electronmicroscopy, the efficacy of a single intratesticular injection ofa chemical agent a sterilize for male rats. The objective of thepresent study was to evaluate, sperm count, sperm motility,sperm morphology, DNA integrity and Teratozoospermia Indexwere evaluated after intratesticular injection of Zinc Oxide Nanoparticlesin rats.Materials and Methods: Sixty adult male Wistar rats weredivided into ۶ groups randomly. After ۱۰ days of acclimationtime, the animals were sedated by xylazine, ketamine cocktail.The testicle diameter related volume of Zinc Oxide Nanoparticles(with different concentrations of ۵, ۱۰ and ۲۵ mg/ml), zincgluconate (۲۵ mg/ml) and sterile distilled water were injectedvia the intratesticular method in each group separately. Rats onexperimental and control groups were killed under anestheticconditions after ۳۰ and ۶۰ days. sperm count, sperm motility,sperm morphology, DNA integrity and Teratozoospermia Indexwere measured.Results: The sperm count, sperm motility, sperm morphologyin Zinc Oxide Nanoparticles group were significantly lowerthan the control group (P<۰.۰۵). On the other hand, DNA integrityin Zinc Oxide Nanoparticles group was significantly higherthan zinc oxide and distilled water groups (P<۰.۰۵). Also, TeratozoospermiaIndex was significantly increased in Zinc OxideNanoparticles group (P<۰.۰۵).Conclusion: Low cost, ease of use, and cultural acceptance ofa castration technique that does not require removal of the testesmake Zinc Oxide Nanoparticles a valuable option for largescaleuse in animals, particularly in remote locations lackingsophisticated clinical facilities or skilled surgeons and staff.Notably, Intratesticular injection of Zinc Oxide Nanoparticlessolution impaired spermatogenesis in rats and has great potentialas a permanent sterilize in this species.
