Evaluation of The Effects of N-Acetyl-Cysteine on Sperm Quality, Chromatin Integrity and Levels of The AntioxidantEnzyme in Infertile Men

چکیده مقاله:

Background: To evaluate effects of supplementation with Nacetyl-cysteine (NAC) on the sperm parameters and biochemicalfactors in terato-asthenozoospermia production of reactiveoxygen species (ROS) is a normal physiologic event in variousorgans, but excessive production of ROS can disrupt spermfunction ,lipid peroxidation, loss of motility and sperm DNAdamage.Materials and Methods: The study was carried out in Unitof Infertility Research Center of the ACECR (Qom. Iran). Thisstudy was performed on ۵۰ infertile men with terato- asthenozoospermia.The patients were given NAC (۶۰۰ mg/d orally)for ۳ months; the control group before received NAC. The semenanalysis was performed according to WHO (۲۰۱۰), protaminecontent [chromomycin A۳ (CMA۳)], DNA integrity [terminaldeoxynucleotidyl transferase-mediated dUTP nick-endlabeling (TUNEL)] assessed. TAC (total antioxidant capacity)and MDA (Malondialdehyde) as stress oxidative markers weredetermined with by ELIZA kit.Results: Percentage of abnormal semen parameters, protaminedeficiency, DNA fragmentation were significantly decreased inpatients was given NAC. At the end of study, the total spermcount, sperm motility and normal morphology in patients weresignificantly higher than before NAC treatment. TAC was significantlyincreased but MDA lower than NAC receiving, Datashowed a negative correlation between TAC and MDA levelsin infertile men.Conclusion: According to the results, medical therapy of terato-asthenozoospermia with NAC supplement could improvequality of semen parameters and chromatin integrity. High degreeof correlation between sperm parameters and antioxidantenzymes suggests reactive oxygen species resulting in reducedsperm quality and levels of antioxidant enzyme.
