Background: Breast cancer is one of the most common canceramong women. Due to lack of the highly specific biomarkers of carcinogenesis and prognostic evaluation, the overall survivalof breast cancer is still dismal. DOK۷ is type of genes whichinvolves in neuromuscular synapses. Researchers reported thatthe expression and methylation of DOK۷ as a tumor suppressor.Also they demonstrated methylation of DOK۷ as an indicatorof breast cancer risk that overexpression correlated with patientsurvival.T he aim of this study was to evaluate the expressionand methylation of DOK۷ gene in breast cancer patients comparedto normal patients and to evaluate its role as a prognosticand diagnostic factor in breast cancer and to investigate the possiblerelationship between DOK۷ gene expression variation andage and methylation status.Materials and Methods: In this study, ۸۰ samples of venousblood were prepared from Farda’s lab to determine the geneexpression and the status of promoter methylation .The sampleswere from the year ۱۳۹۵.The expression of DOK۷ gene was investigatedby Real-Time PCR and DOK۷ promoter methylationwas investigated by MS PCR.Results: In this study, we found that the expression of DOK۷was decreased in patients’ blood samples compared with normalblood samples and the reduced expression is associatedwith promoter methylation and also the results of DOK۷ promotermethylation showed that ۷۲.۵% of promoter in patients’blood samples was methylated and ۲۰% of promoter in normalblood samples was un methylated.Conclusion: The reduced expression of DOK۷ and DOK۷ promotermethylation in patients’ blood samples would be a candidatebiomarker for breast cancer prognosis.
