Cardiomyocyte Differentiation of WhartonʼS Jelly Mesenchymal Stem Cells by Different Cocktaies: Based On۵-Azacytidine

چکیده مقاله:

Background: Many studies have shown the ability of Wharton'sJelly Mesenchymal Stem Cells (WJMSCs) to differentiateinto cardiomyocyte-like cells under various inductions. ۵-azacytidine(۵-aza) induces cardiomyocyte differentiation by activatingextracellular regulated kinases. In the present studies wehave investigated the effect of different cocktails of ۵-aza forcardiomyocyte differentiation of WJMSCs.Materials and Methods: WJMSCs were harvested by explantculture method and mesenchymal surface markers (CD۳۴,CD۴۴, CD۴۵, CD۷۳, CD۹۰ and CD۱۰۵) were characterized byflow cytometry of mesenchymal cell markers. Also, adipogenicand osteogenic differentiation was evaluated. For cardiomyocytedifferentiation, ۴ cocktails including: ۱) [۵-aza], ۲) [۵-aza+ ascorbic acid (Asc)], ۳) [۵-aza + hypoxia (Hypo)], ۴) [۵-aza+ Asc + Hypo] were examined. The expression of Connexin۴۳,GATA-۴ and troponin I (cTnI) was evaluated by real time PCR.Results: The isolated spindle like cells did not express hematopoieticlineage markers such as CD۳۴ but expressed CD۴۴,CD۴۵, CD۷۳, CD۹۰ and CD۱۰۵. Also, the cells exhibited osteogenicand adipogenic differentiation ability. The expressionof GATA-۴ and cTnI was higher in [۵-Aza + Asc] group, withGATA-۴ as the significant cardiogenic marker. Connexin۴۳ hadhigher expression in [۵-Aza + Hypo] group without any significantdifference.Conclusion: GATA is one of the first implicated transcriptionfactor in the regulation of cardiomyocyte differentiation.As GATA and cTnI had higher expression level in [Aza + Asc]group, we may conclude that this cocktail can affect cardiomyocytedifferentiation. We also may conclude that ۵-azacytidinealong with ascorbic acid or hyposia are more suitable inducersfor cardiomyocyte differentiation than ۵-azacytidine alone and(Aza + Asc) is bether than (Aza + Hypo).
