Background: The epigenetic memory of a cell is a set of modificationsto the cell deoxyribonucleic acid (DNA) that do notalter the DNA sequence. These modifications can alter gene expression.Induced pluripotent stem cells (iPSCs) have been generatedfrom various somatic cell types. Many study have beensuggested that iPSCs from different cell sources have distinctmolecular and functional properties including self-renew differentiateinto any specialized cell of the body. Although, iPSCscan make almost varied cells, there are concerns about variationin the differentiation capacity of these pluripotent cellscompared to ESCs. It would be useful that we detect the majorsources of variation for their use in clinical applications anddisease modeling. To uncover these variations, we have investigatedhistone methylation marker during direct differentiationof iPSCs and ESCs into Neural cells.Materials and Methods: We have used data set to address theissue with differential capacity among ES and iPSCs. We obtaineddifferentially expressed (DE) genes. Next, methylationpattern of promoter regions of these DE genes was assessed.Then, differentially methylated regions (DMR) that are involvedin defining differential capacity were extracted.Results: Our analysis shows that expression patterns of somegenes are different. These data identified DNA methylation asan essential epigenetic mechanism and determines stem cellfunctions. Additionally, affected DMR regions causing differentialgene expression pattern showed promising results.Conclusion: Here we demonstrate that ESCs and iPSCs havean epigenetic memory of their tissue of origin and it had led todifferent gene expression patterns. These analyses revealed theprincipal differences between iPSCs and ESCs. We hope theseanalyses would be beneficial for applications in disease modellingor treatment.
