Neuroprotective Effect of Mesenchymal Stem Cells Derived from Human Embryonic Stem Cell -ConditionedMedium on Ischemic Stroke Model

Background: Stem cell therapy is a promising approach forstroke. Although low survival rates and potential tumorigenicityof implanted cells could reduce the efficacy of the cell-basedtreatment, recent investigations have proven that the use of stemcell-conditioned medium (CM) may be a feasible approach toovercome these limitations. There are numerous cytokines andgrowth factors in the CM of various stem cells responsible forthe paracrine protective effects of stem cells. The purpose ofthe present study was to assess the effect of the conditionedmedium of mesenchymal stem cells derived from human embryonicstem cell on infarct volume and neurological functionsin ischemic stroke model rats.Materials and Methods: Ischemic stroke was inducedby standard right middle cerebral artery occlusion method(MCAO) in the ۸-week old wister male rats. Injection of theCM or DMEM (۵μl) was respectively done into the left lateralventricle of the treatment and control animals from one hourfollowing the surgery to the second day after MCAO induction(three doses). Behavioral tests including the cylinder and modifiedneurological severity score (mNSS) were performed on ۱,۳ and ۷ days after the injury. Infarct volumes were evaluated on۳ and ۷ days after the MCAO by the ۲,۳,۵ triphenyltetrazoliumchloride (TTC) staining method.Results: Our results indicated that ۹۰ min occlusion of the arterycaused impairments in sensory-motor functions in the ischemicrats. Furthermore, i.c.v injection of the CM improvedfunctional recovery in the treatment group after the stroke. Volumeanalysis revealed that animals subjected to treatment withCM had significantly smaller infarcts than controls.Conclusion: In this study, the therapeutic effects of the CMagainst stroke were confirmed in an animal model.
