Enrichment analysis of potential hub genes in Acute Myeloid Leukemia based on TCGA datasets

Acute myeloid leukemia (AML) is the most common acute leukemia in adults. AML is amalignancy of the stem cell precursors of the myeloid lineage (red blood cells, platelets, and whiteblood cells other than B and T cells). Like other malignancies, it is due to genetic variations that lead toneoplastic changes and clonal proliferation. Despite great advances in the diagnosis and treatment ofcancer, the treatment has big challenges. Nowadays, with the advances in the bioinformatics analysisof RNAseq, it can identify potential biomarkers for diagnosis, targeting treatment, and evaluation oftumor metastasis and relapse. In the current study, we conducted the gene ontology and Kegg pathwaybased on TCGA datasets.At first, we used GEPIA۲ to examine the TCGA AML dataset to identify all DEGs linked with AMLamong high throughput RNA-Seq data. After analyzing the survival data of AML, a Protein-proteininteraction (PPI) network of significant genes associated with AML was constructed in Cytoscapesoftware, and the hub genes were identified. for enrichment analysis, we used the Enrichr website(https://maayanlab.cloud/Enrichr/) and extracted the Kegg pathway and gene ontology based on thepotential hub GeneTwelve Hub genes (MRPS۱۰, MRPL۱۶, NDUFS۸, MRPL۴, ENO۱, MRPL۱۲, MRPL۲, EPRS۱,SDHA, SOD۱, HSPA۵, SDHB) were extracted from PPI based on their degree. the result showed in the۱۵۷ biological processes, the Mitochondrial Translation (adj-P-value=۱.۳۲E-۰۷), Translation (adj-Pvalue=۱.۳۲E-۰۷) and Mitochondrial Gene Expression (adj-P-value=۱.۳۲E-۰۷) was significantlymeaningful. In ۲۶ cellular components, we identified just ۳ components with meaningful adj p-valueswhich included Mitochondrial Inner Membrane (adj-P-value=۱.۱۱E-۰۸), Organelle Inner Membrane(adj-P-value=۱.۱۱E-۰۸) and Mitochondrial Membrane (adj-P-value=۶.۱۴E-۰۸). In ۳۴ molecularfunctions, RNA Binding (adj-P-value=۰.۰۰۲۶۴۵۳۷۶) and GTPase Binding (adj-P-value=۰.۰۰۴۸۷۴۷۸۹)have a more meaningful. At the last, in the ۲۵ Kegg pathway, we identified Ribosome (adj-Pvalue=۵.۴۷E-۰۷) with meaningful P-value.This study identifies gene ontology and keg pathways of hub genes which involved in the occurrenceand progression of Acute Myeloid Leukemia. This information may hold promise as potentialbiomarkers and therapeutic targets.
