Identification of crucial genes and pathways associated with Hepatocellular carcinoma based on bioinformatics analysis

Among the various forms of liver cancer diagnosed in ۲۰۲۰, hepatocellular carcinoma(HCC) took the lead, constituting approximately ۷۵-۸۵% out of the nearly ۹۰۶,۰۰۰ reported cases.Additionally, HCC ranked as the third highest cause of cancer-related deaths globally in ۲۰۲۰, with anestimated ۵-year survival rate of ۲۰%. The primary aim of this study is to ascertain novel biomarkersassociated with HCC, enabling the prediction of prognosis and the advancement of treatment strategies[۱,۲].Methods: The GSE۱۰۱۶۸۵ dataset was obtained from GEO database, consisting of ۲۴ cancer and ۸normal samples. To assess the consistency of the samples, Principal Component Analysis (PCA) wascarried out. Subsequently, the statistical analysis was performed utilizing the LIMMA package.Differential expression of genes (DEGs) was determined based on the criteria of |𝑙𝑜𝑔۲𝐹𝐶|> ۳ and adjP-value < ۰.۰۵. To examine the relationship between gene expression and survival outcomes, theKaplan-Meier plotter was utilized.Results: Microarray analysis was conducted to assess the expression profile of ۶۹ samples. A total of۱۲۶ DEGs were identified, of which ۴۳ were up-regulated and ۸۳ were down-regulated. Among them,two genes SLCO۱B۳, and CLEC۴M have been discovered in previous studies to be related to cancerssuch as Prostate and lung. These two genes show a significant decrease in expression in this dataset.Through the utilization of the GeneCards and Reactome pathway databases, it has been determined thatthese genes possess the ability to regulate significant pathways, including Bile acid and bile saltmetabolism and Complement cascade. According to Kaplan-Meier plots DEGs are correlated with poorprognosis in HCC.Conclusion: Our examination reveals that the SLCO۱B۳ and CLEC۴M genes are vital in HCC andstrongly associated with poor prognosis.
