The role of imatinib on the differentiation genes expression in K۵۶۲ cell line: A bioinformatic study

چکیده مقاله:
Bcr-Abl protein is a tyrosine kinase enzyme that is involved in the growth and carcinogenesisof K۵۶۲ cells in chronic myeloid leukemia cancer [۱, ۲].Therefore, the inhibitors of this enzyme suchas imatinib can reduce the growth of K۵۶۲ cells[۲] . In this study, the aim of this work was to investigatethe role of imatinib in the expression of the differentiation genes of K۵۶۲ cells into red blood cells [۳].In this study, the GEO site the GEO۲R tool were used to find and analyze the dataset. After surveyingthe site, and checking the different datasets, the GSE۱۹۵۶۷ dataset was chosen to analyze the imatinibeffect on K۵۶۲ cell line gens expression profile[۴]. Analysis of the GSE۱۹۵۶۷ dataset displayed overexpressionof ALAS۲, HBB, HBD, GYPA, FECH, TAL۱ genes in imatinib treated K۵۶۲ cells by ۲۲.۳,۱۱.۷, ۱۰.۷, ۴.۲, ۳.۰۱, and ۲.۴ times, respectively. The results showed that the expression of genesinvolved in the metabolism of heme and hemoglobin and also Glycophorin gene has been increased intreated cells. According the role of the genes in the differentiation of the K۵۶۲ cells, it can be suggestedthat imatinib induces differentiation in the cells. It may be related to inhibition the Bcr-Abl proteinwhich causes K۵۶۲ cell growth.
