Investigating antioxidant activity, total phenolic and total flavonoid contents in root and leaf of transgenic medicinal plant Withania somnifera by Inplanta transformation method

One limitation of using tissue culture to produce hairy roots to increase metabolite production is the high risk of contamination and the time-consuming nature of the process. The Inplanta transformation method was used in this study to address this issue. In this method, the Agrobacterium rhizogenes bacterium is directly injected into the primary node of growing seedling plants. Specifically, the A۴ strain of bacteria was injected into the primary node of two-leaf stage Withania somnifera seedlings. Hairy roots developed after ۱۵ days. The transgenic plants developed from the hairy roots were then grown under controlled conditions of ۲۵°C temperature and ۶۰-۷۰% humidity. The amounts of phenols, flavonoids and antioxidant properties were analyzed in the roots and leaves of the transgenic plants and compared to control plants. The experiment followed a completely randomized design with three replications. Statistical analysis was performed using SPSS version ۲۲ software and graphs were generated in Excel. Analysis of variance results showed significant differences between treatments for all traits studied. No significant differences were observed between the transgenic and control plants in terms of changes in phenolic and flavonoid compounds in the leaves and roots. However, the leaves of the transgenic plants showed higher antioxidant activity (DPPH) than the control plants.
