Study of Essential Oil and Effect of Temperature and Seed Appendages (pappus) on Germination Characteristics in Coltsfoot

Coltsfoot (Tussilago farfara L.) is a perennial plant of the Asteraceae family that has been used as an ancient folk remedy to treat coughs, bronchitis, and asthmatic disorders. Ripe seeds were collected from T. farfara plants growing in Pol-e Zangholeh, Iran, in this study. Samples were gathered by the Tarbiat Modares University (TMU) Department of Horticultural Sciences laboratory in late April ۲۰۱۶. The extracted seed essential oil constituents identified and quantified using GC/MS and GC, respectively. Phy,tol, n-Nonadecane, n-Tetradecane, and ۴,۴-Dimethyltetracyclo [۵.۲.۱.۰۲,۶.۰۳,۵] decane, with amounts of ۳۰.۵%, ۱۱.۴%, ۹.۸%, and ۶.۲%, respectively, were the main components in the seeds oil of T. farfara. The germination variables were studied using a factorial experiment with ۵ replicates × ۵۰ seeds in a totally randomized design. Treatments include: cultivating non-pappus-bearing seeds in a Petri dish and placing them at ۴°C (c۱t۱) and ۲۵ °C (c۱t۲), Cultivating pappus-bearing seeds in a Petri dish and keeping them at ۴ °C (c۲t۱) and ۲۵ °C (c۲t۲) and cultivating of non-pappus-bearing seeds treated with sodium hypochlorite ۱.۵% for ۲۰ minutes and keep them at ۴ °C (c۳t۱) and ۲۵ °C (c۳t۲). The interaction between culture type and temperature was significant (p≤۰.۰۱) for the germination percentage of seeds, germination rate, and seed vigor of T. farfara, according to analysis of variance. The rising seed germination rates of C۳t۲ and C۳t۱ were reported at ۷۶.۴% and ۷۴.۸%, respectively. The results revealed the closest positive correlation between germination rate, vigor index, and seed germination percentage (p≤۰.۰۰۱).
