Laminin γ, High Mobility Group Box I (HMGB۱), and Collagen Type-۱ for Detection of Enterococcus Faecalis Microbial Surface Components Recognizing Adhesive Matrix Molecules (MSCRAMMs)

The inflamed pulp can develop into pulp necrosis and requires endodontic treatment. The failure of endodontic treatment is mainly caused by Enterococcus faecalis infection. The mechanism is mediated by the ability of E. faecalis, as a gram-positive bacterium, to form a biofilm covering the cell membrane, which is known as microbial surface recognizing adhesive matrix molecules (MSCRAMMs). The aim of this study was to identify MSCRAMMs through the expression of laminin γ, High Mobility Group Box I (HMGB۱), and type-۱ collagen. All groups were prepared for molar cavitation using a round bur and injected with E. faecalis bacteria with a concentration of ۱۰۰%. They were observed for ۲۴ hours, ۷۲ hours, and ۱۲۰ hours after injection. The K group was the control group containing rats that were not received a bacterial injection at ۲۴ hours, ۷۲ hours, and ۱۲۰ hours. After that, the tooth was decalcified using ۱۰% EDTA at ۳۷ °C, which took up to two months. Macrophages were observed using a light binocular microscope. Significant results (p<۰.۰۵) were found in laminin γ and HMGB۱ between the control with ۲۴ hours groups, also insignificant results (p>۰.۰۵) were found between ۲۴ with ۷۲ hours group and ۷۲ with ۱۲۰ hours group. All groups within the collagen type-۱ group were not significant. Both laminin γ and HMGB۱ are good parameters to predict Enterococcus faecalis growth phase and biofilms production with the log phase happening before ۲۴ hours and the stationary phase happening after ۲۴ hours.
