Rectal cancer, a highly lethal subtype of colorectal cancer, poses significant challenges intreatment and prognosis due to its complex molecular and genetic heterogeneity. This studyleverages advanced RNA sequencing (RNA-seq) and systems biology approaches to identify keymolecular factors involved in rectal cancer. By integrating transcriptomic data from primarytumor samples and adjacent normal tissues, we identified differentially expressed genes (DEGs)and their associated signaling pathways. The IL-۱۷, Wnt, and cytokine-cytokine receptorinteraction pathways emerged as critical in rectal cancer progression. Protein-protein interactionnetwork analysis highlighted CXCL۱, CXCL۸, and MMP۱ as hub genes with significant roles intumor development. Kaplan-Meier survival analyses were performed to assess the prognosticvalue of these genes. Interestingly, while molecular studies suggest significant roles for thesegenes in cancer progression, survival analyses showed no statistically significant differences inoverall survival based on their expression levels. MMP۱ exhibited a trend towards improvedsurvival with high expression, although not reaching statistical significance.Despite the lack ofclear prognostic value, our molecular pathway analysis underscores the complex interplay ofthese genes in promoting rectal cancer through various mechanisms, including angiogenesis,epithelial-mesenchymal transition, and extracellular matrix remodeling. This study highlights thepotential of CXCL۱, CXCL۸, and MMP۱ as therapeutic targets and suggests that combinationtherapies targeting multiple pathways may be more effective in treating rectal cancer. Ourfindings contribute to a deeper understanding of rectal cancer biology and pave the way forfuture research into more personalized and effective treatment strategies.
