چکیده مقاله:
Metastatic melanoma is a challenging form of cancer that has traditionally been difficult to treat.Recent advancements in immunotherapy have revolutionized treatment options, leading to increasedsurvival rates for patients. However, the rapid development of new treatments has also introducednew challenges, including severe side effects and drug resistance. In this study, a novel peptidedelivery system called KK-۱۱b was developed to increase the targeted delivery of achemotherapeutic drug, doxorubicin, to melanoma cells. The stability of KK-۱۱b was analyzed, andpreliminary in vitro studies showed promising results. Further research will focus on the cytotoxicityof the KK-۱۱b conjugate in different types of melanoma cells and in the presence of human serum,with the goal of progressing to in vivo murine studies.Method:The ligand KK-۱۱b was synthesized to form a cell-specific, noncytotoxic peptide residue and thenconjugated to a linker, sulfo-SMCC, and doxorubicin. The resulting peptide-drug conjugates (PDCs)were characterized for stability and tested in preliminary in vitro melanoma cell studies.Result:KK-۱۱b demonstrated stable presence in serum-free cell media, water, and in preliminary in vitroA۳۷۵ melanoma cell studies. Further analysis is needed to determine the cytotoxicity of the KK-۱۱bconjugate in other melanoma cell types and in the presence of human serum.Conclusion:The development of KK-۱۱b as a targeted drug delivery system shows promise for improving thetreatment of metastatic melanoma. Further research is necessary to fully understand the potential ofPDCs in treating this aggressive form of cancer.
