Evaluation of the Interaction between Ferulic Acid and RNA via Comprehensive Multispectral Methods and Molecular Docking Studies

The interaction between ferulic acid (FA) and Ribonucleic acid (RNA) under physiological conditions (Tris-HCl buffer solutions, pH ۷.۴) was investigated by UV-Vis spectroscopy, fluorescence spectroscopy, and molecular docking. Results indicated that a complex of ferulic acid with RNA was formed with a binding constant of Kb (۲۹۸.۱۵K) = ۱.۰۲×۱۰۳ M-۱. Negative value of Gibb’s-free energy change revealed that the process is spontaneous. The thermodynamic parameters such as enthalpy change (ΔH۰) and entropy change (ΔS۰) showed that the acting forces between FA and RNA mainly included hydrophobic interactions. The obtained theoretical results were in good agreement with those obtained from experimental studies. The knowledge gained from this study will be helpful to further understand the RNA binding mechanism and can also provide much fruitful information for designing a new type of anticancer drugs.
