Breast cancer is the leading type of invasive cancer among women globally. Despite treatmentadvancements, it continues to be a significant cause of illness and death. Breast cancer treatment has madesignificant progress, but it still faces challenges. Recently, alternative medicine has gained attention as apromising option due to its availability, affordability, effectiveness, and minimal side effects. Herbal phenoliccompounds, such as apigenin, oleuropein, and hydroxytyrosol, found in olive leaf extract, are being consideredas potential alternatives for cancer treatment. These compounds possess antioxidant and antiproliferativeproperties, making them promising candidates for various diseases including cancer, bacterial infections,inflammation, diabetes, neuroprotection, cardiovascular health, and liver protection. The present study aims toinvestigate the in vitro anticancer effects of olive leaf extract on human breast cancer cell lines. We used anultrasound approach to powder olive leaves and obtain their phenolic extract, which was then characterizedusing HPLC, The IC۵۰ of olive leaf extract on breast cancer cells was found to be ۸۷ ± ۰.۵ μm/ml after ۷۲ h, invitro anticancer activity was demonstrated by monitoring cell morphology with DAPI staining, which showed thecharacteristics of apoptosis and nuclear abnormalities. In addition, the molecular anti-cancer investigation ofthe extract was performed by analyzing the expression of key genes of the cancer pathway. The study foundthat olive leaf extract had a strong, dose-dependent effect on inhibiting the growth of MCF-۷ breast cancer cells.It was also found to have antioxidant effects on the cells. These results suggest that olive leaf extract could be apromising supplement for breast cancer treatment.
