Apigenin attenuates serum concentrations of TNF-a, interleukin ۱b and interleukin ۶ in lipopolysaccharide-stimulated rats

Objective: The use of flavonoids is increasing due to their costeffectiveness and less adverse reaction. Therefore, the effect ofapigenin on lipopolysaccharide (LPS)-induced inflammation wasinvestigated by measuring IL-۱b, IL-۶, and TNF-a, of serum in themale rats.Materials and Methods: Ninety male wistar rats were divided in۶ groups included; control, sham, dexamethasone ۱۵ mg/kg,intraperitoneally (i.p.), and apigenin (۵, ۱۵, and ۳۰ mg/kg, i.p).Thirty minutes after the administration of solvent or apigenin, LPS(۳۰ μg/kg, i.p) was injected. At time intervals of ۴, ۱۲ and ۲۴ hrafter injection, blood samples were taken and the concentrations ofTNF-a, IL-۱b and IL-۶ were measured by enzyme-linkedimmunosorbent assay.Results: Compared to the control, apigenin (۵ mg/kg) decreasedthe level of TNF-a, and IL-۱b in a period of ۲۴ hr (p<۰.۰۵). Theconcentration of IL-۶ decreased significantly by apigenin (۱۵mg/kg) ۲۴ hr after injection (p<۰.۰۵). Apigenin (۳۰ mg/kg)decreased the level of TNF-a, at all three time points (۴ hr; p<۰.۰۵,۱۲ hr; p<۰.۰۱, and ۲۴ hr; p<۰.۰۱), and the level of IL-۱b (p<۰.۰۱),۲۴ hr and the level of IL-۶ at ۴ hr (p<۰.۰۵), and ۲۴ hr (p<۰.۰۱)after LPS injection.Conclusion: Apigenin can suppress serum inflammatorycytokines, similar to dexamethasone
