Alzheimer´s disease (AD) is a neurodegenerative disorder without any specific cureand rapid rise in incidence. AD is the most common cause of dementia globally, and it isreported that approximately ۵۰ million people are suffering from dementia, globally. Riskfactors for AD occurrence comprise different factors such as family, age, and some particulargenes comprising Apolipoprotein E (ApoE). Apolipoproteins (Apo) are a group ofamphipathic molecules, that existing as the main components of plasma lipoproteins. ApoElipoprotein particles loaded with lipids bind to different receptors of the cell surface tosustenance membrane homeostasis and injury repair in the human brain. ApoE can be foundin three main alleles, ApoE۲, ApoE۳, and ApoE۴. ApoE۳, a high-density lipoprotein, is themost usual isoform. ApoE۳ is individualistically essential for neurovascular function, and it isspecifically involved in AD, producing complex effects including alternations in clearanceand aggregation. Technological advancements through the past years have implicated ApoEin other pathophysiological aspects of AD. Many ApoE-directed therapeutic approaches havebeen shown to be useful in mouse models as a potential target for translation to humanclinical studies.In this review, our main focus is on ApoE۳ allele interactions with AD, and its indirect rolesthat affect some features in AD models.
