Long Non-coding RNA FEZF۱-AS۱: A Potential Biomarker in Colorectal Cancer

Introduction: Colorectal cancer (CRC), a member of the quintet of most widespread and fatal malignancies,presents a complex interplay of factors originating from environmental exposures, dietary patterns, age, andgenetic predisposition. While significant strides have been taken in understanding the etiology of CRC, thespecific genetic determinants fueling its pathogenesis are not fully delineated. The long non-coding RNA(lncRNA) FEZF۱-AS۱ has demonstrated its influence on human cancers, notably colorectal cancer. Hence, theprimary aim of this systematic review was to scrutinize the potential of FEZF۱-AS۱ as a biomarker in colorectalcancer.Search Method: An extensive search of the PubMed, Scopus, and Web of Science databases using thekeywords “FEZF۱-As۱” and “colorectal cancer” from ۲۰۱۳ to ۲۰۲۳ yielded ۹۴ articles. Following theapplication of inclusion and exclusion criteria, a total of ۱۰ articles were chosen for comprehensive review.Conclusion: Among the ۱۰ articles focusing on the role of FEZF۱-AS۱ in colorectal cancer, ۷ demonstratedthat reduced levels of FEZF۱-AS۱ suppressed CRC cell migration, invasion, and proliferation, and inhibitedtumor growth in a CRC nude mouse model, while ۳ did not provide evidence of such an association.Additionally, it was observed that FEZF۱ enhances the levels of cytoplasmic and nuclear protein kinase andpyruvate kinase (PKM۲), thereby influencing colorectal cancer progression via PKM۲ activities. These findingsdemonstrate the involvement of FEZF۱-AS۱ in the development and progression of CRC. The expression ofmiR-۶۳۲ was significantly reduced in rectal cancer cell lines, and inhibiting miR-۶۳۲ partially reversed theinhibition of rectal cancer cell growth, invasion, and migration caused by FEZF۱-AS۱ knockdown. Furthermore,the results suggest that the FEZF۱-AS۱/miR-۶۳۲/FAM۸۳A axis plays a role in promoting the progression ofrectal cancer. In conclusion, FEZF۱-AS۱ can serve as the potential diagnostic and prognostic biomarker forCRC patients.
