BACKGROUND AND ABJECTIVEBreast cancer is one of the most common cancers in women in both developed and developing countries, and it is the main cause of cancer-related death among women. And the main cause of death is cancer among women. However, if it is detected in the early stages, it can be treated. Due to the greater understanding of the molecular pathways involved in the pathogenesis of cancers, several macromolecules at the level of RNA, DNA and protein have been investigated as biomarkers in the diagnosis and prognosis of cancers. Therefore, the members of the miR-۲۰۰ family are one of the oncogenic miRNAs in breast cancer that are associated with increased expression, which we will discuss with the clinical and bioinformatics studies of the members of the miR-۲۰۰ family on the target genes of E۳ ubikinase and KRAS in intracellular signaling pathways.MATERIALS AND METHODSAccordingly, the paraffin blocks used for this study include ۳۰ breast cancer paraffin blocks and ۳۰ normal paraffin blocks. The average age of the samples was between ۴۵ and ۶۰ years, and the progress and stage of the disease is in Stage II and Stage III, and then extracting RNA from healthy and normal tissue samples, and then extracting the corresponding miRNA with a special kit and detecting quantitative RNA and cDNA synthesis, and finally measuring the expression level The target genes and miRNAs were analyzed by Real time PCR technique.RESULTS AND DISCUSSIONIn this study, the miR-۲۰۰ family caused the average KRAS gene expression to increase by ۱.۴ times (P<۰.۰۵) in patients with breast cancer compared to healthy individuals by examining the ΔΔCt method, but the level of E۳ gene expression in breast cancer patients Compared to healthy people, it has decreased by ۳.۴ times (P<۰.۰۵).CONCLUSIONFinally, the results obtained from this study Clinical/bioinformatics showed that among the members of the MIR-۲۰۰ family (MIR-۲۰۰b and MIR-۱۴۱) with the aim of placing E۳ and KRAS genes in important biological processes and molecular pathways involved in Breast cancer with expression profile changes in tumor tissue samples can be used as a biomarker. to be diagnostic/prognostic in breast cancer
