Identification of Candida species isolated from oral infection of head and neck cancer patients by PCR RFLP method

BACKGROUND AND OBJECTIVESCandida species are the most important cause of nosocomial fungal infections.Oropharyngeal candidiasis is a common disorder in patients with head and neck cancer thatoccurs during radiotherapy and begins with a post-colonization phase of the oral mucosa.Due to the importance of detecting Candida species, this study was conducted in order toidentify the species by RFLP molecular method to recognize this yeast.MATERIALS AND METHODSOral swab samples were all cultured on Sabouraud Dextrose Agar medium for ۴۸ h at ۲۸ °Cand then sub-cultured on a chromogenic medium. Final identification was confirmed by PCRRFLP method. All polymerase chain reaction (PCR) products underwent digestion byrestriction enzyme MspI. The digested products were electrophoresed on agarose gel ۱.۷%.Each isolate was identified by comparing the obtained band patterns for Candida species.RESULTS AND DISCUSSIONOur results showed that the most frequently Candida species from a total of ۲۳ isolates wasC. albicans (۶۵.۲۱%) followed by C. tropicalis (۲۶.۰۸%) and C. glabrata (۸.۶۹%).CONCLUSIONAmong the ۲۳ isolates isolated from patients with oropharyngeal candidiasis in this study,based on the molecular method, Candida albicans was the most prevalent. Candida tropicalisranks second and Candida glabrata has the lowest prevalence .
