Screening of effective bacteriophages against Pseudomonas aeruginosa from Dasht Desert

BACKGROUND AND OBJECTIVESAntibiotic-resistant Pseudomonas aeruginosa is the common causes of hospital infections, Therefore, bacteriophage therapy is a suitable alternative for the treatment of P. aeruginosa infections. This study aimed to isolate of effective bacteriophages against P. aeruginosaMATERIALS AND METHODSIn this study, soil samples were collected from a garden located in Semnan province. Soil and sterile distilled water were mixed in a ۱:۱ (w/v) ratio and shaken for ۸ hours at ۳۰°C. Then this mixture was centrifuged and supernatant was filtered. The filtered supernatant was mixed with equal amount of nutrient broth, ۲۵ μl of Pseudomonas aeruginosa culture and shaken at ۳۷°C for ۳۶ hours. After centrifugation and filtration, phage lysate was concentrated using NaCl and polyethylene glycol ۸۰۰۰. Plaques morphology and number of phages in concentrated solution, MOI value and stability of phage in different physical conditions were studied by double layer agar method. Transmission electron microscopy (TEM) was used to determine the morphology and phage family.RESULTS AND DISCUSSIONPhage plaques produced spherical shape with a semi-clear halo around the center. The number of bacteriophage in the phage concentrate was equivalent to ۳.۵ × ۱۰۱۱ PFU/mL, the MOI value is ۸.۷۵ and most stable at ۴°C and pH ۷. The morphology analysis with TEM, showed that phage had an icosahedral head with a contractile tail about ۱۴۰ nm that belongs to the myoviridae family. In ۲۰۲۱, Lashtoo Aghaei et al isolated eighteen different bacteriophage against Pseudomonas aeruginosa from sewage and showed that bacteriophage were most effective in lysing Pseudomonas aeruginosa. In ۲۰۲۲, Abo Kamer et al isolated and identified a new bacteriophage against Pseudomonas aeruginosa and evaluated its potential effect in the treatment of skin infection.CONCLUSIONThe results of these preliminary investigations indicate the bacteriophages may be considered for use in phage therapy against Pseudomonas aeruginosa.
