Studying the survival of epithelial cells in the presence of native probiotic strains

BACKGROUND AND OBJECTIVESProbiotics have attracted a lot of attention due to their potential therapeutic applications and the production of various metabolites such as vitamins. In this study, the survival of epithelial cells in the presence of ۷ probiotic strains and the production of some B vitamins were investigated.MATERIALS AND METHODSIn order to measure the viability, A۵۴۹ cells were added to the number of ۱۰۴ × ۴.۵ cells in each well of the ۹۶-well plate. Next, the cell sediment of probiotic bacteria was washed thrice with sterile phosphate buffered saline and added to the culture medium containing metabolites of probiotics with cell dilution series of ۱۰۸, ۱۰۶ and ۱۰۵ cfu/ml. After ۲۴ hours of incubating, the survival of epithelial cells were studied by MTT assay. To measure vitamin production by probiotic strains, ۲۰ μl of culture medium containing metabolites of probiotics and solution of vitamins B۲, B۳ and B۶ were coated on chromatography paper and immersed in the mobile phase for separation. Chromatograms were developed and, after drying, viewed under a UV lampRESULTS AND DISCUSSIONThe results showed that eukaryotic cells have a potential to survive in the presence of ۱۰۵ dilution of probiotics in the range of ۴۰ to ۱۰۰%, and survival is directly correlated with the quantity of bacteria. SUBC۴ and SUBC۲ strains had the lowest and highest survival rates in dilution ۱۰۸, with ۷۵% and ۱۰۰%, respectively. As Song et al. studied effect of probiotics on gastric epitelial cells in ۲۰۱۹. Additionally, in TLC, the results of calculating the Rf value, revealed that probiotic strains produce the vitamins B۲, B۳, and B۶. As Indira, M., et al. studied bioactive molecules produced by probiotics at ۲۰۱۹.CONCLUSIONA promising strategy for advancing both health and innovation in the food industry is the inclusion of probiotics in food products.
