The inhibitory effect of Lactobacillus rhamnosus on p۳۸-MAPK pathway activation and the expression of IL-۱۷A

BACKGROUND AND OBJECTIVESMultiple sclerosis (MS) is one of the most common autoimmune disorders of the central nervous system (CNS), characterized by excessive immune cell infiltration, degeneration of the myelin sheath, and apoptosis of neuronal cells. It is now known that inflammation significantly contributes to the pathogenesis of the disease. Several lines of evidence have demonstrated that the p۳۸ mitogen-activated protein kinase (MAPK) pathway regulates the immune cell response and programmed cell death and cell survival. Also, it has been shown the activation of the p۳۸-MAPK pathway participates in the pathophysiology of various neurodegenerative disorders, such as Parkinson’s disease and MS. In the present study, we examined the effect of Lactobacillus rhamnosus on the expression of p۳۸-MAPK and Interleukin-۱۷A (IL-۱۷A) in a murine model of MS called experimental autoimmune encephalomyelitis (EAE).MATERIALS AND METHODSMice were immunized with myelin oligodendrocyte glycoprotein ۳۵-۵۵ (MOG۳۵-۵۵), followed by the injection of pertussis toxin to induce paralysis EAE mice. Following the induction of EAE, the animals received Lactobacillus rhamnosus by gavage when the early clinical signs of EAE began to appear. ELISA and real time pcr was done for expression of p۳۸-MAPK and IL-۱۷A.RESULTS AND DISCUSSIONThe results showed that the administration of Lactobacillus rhamnosus led to a noticeable reduction in the clinical score of EAE mice. Moreover, the protein expression of the phosphorylated form of p۳۸-MAPK was significantly diminished in the sacral region of the spinal cord of EAE mice receiving Lactobacillus rhamnosus when compared with EAE mice receiving no treatment (control group). The real-time PCR technique indicated that Lactobacillus rhamnosus lowered the expression of IL-۱۷A in the spinal cord of EAE mice compared with the control group.CONCLUSIONFinally, it seems that probiotics, such as Lactobacillus rhamnosus, could be used as an alternative therapeutic option for alleviating neurological diseases in which the p۳۸ MAPK signaling pathway is involved.
