BACKGROUND AND OBJECTIVESArginase deiminase is an enzyme that breaks down arginine. Arginine is an essential amino acid for the growth of cancer cells. This enzyme is used to treat Alzheimer's disease and also to treat some cancers such as hepatocellular carcinoma, melanoma, and mesothelioma. Due to the wide application of this enzyme in industry and medical sciences, we produced this enzyme in a recombinant form.MATERIALS AND METHODSIn this research, the arginase deiminase enzyme gene was cloned into the pet۲۱ vector, then the PCR technique was used to confirm the presence of the desired gene in the bacteria. Then it was transformed by the heat shock method in Escherichia coli strain BL۲۱ and using IPTG with different concentrations and temperatures. The expression of this enzyme was optimized.RESULTS AND DISCUSSION To confirm the transformation process, first, the growth of bacteria was checked in the culture medium containing kanamycin and the positive transformation samples grew, unlike the negative transformation samples. PCR results also indicate the success of cloning and enzyme transformation. After confirming the presence of the arginase deiminase gene in Escherichia coli strain BL۲۱, the results showed proper protein expression.CONCLUSIONSince the enzyme arginase deiminase was produced in a good way. Therefore, in the next steps, it can be produced on a larger scale to be used for therapeutic purposes.
