The Study of antibacterial effects of five Iranian plant Methanolic extracts on Helicobacter pylori isolates Resistance in East Azerbaijan province

BACKGROUND AND ABJECTIVEHelicobacter pylorus is considered as an important human pathogen which is associated with a variety of gastrointestinal diseases, such as chronic gastritis, peptic ulcers, stomach cancer and MALT lymphoma. So,H. pylori eradication is recommend being the most effective method for healing gastric ulcers. In fact, the main reason for failure was recognized to be H. pylori resistance to antibiotics. Accordingly, the purpose of this research was to study the antibacterial effects of five Iranian plants on H. pylori isolates resistance to metronidazole.MATERIALS AND METHODSA biopsy specimen was obtained from each of ۱۰۰ or ۲۶۸ patients with dyspepsia referred to hospital. Then, they were transferred to the laboratory in transport medium (۱% agar). The biopsies were first defibrinate din Brucella agar medium containing ۷% sheep blood, and vancomycin (۵mg/L), polymyxin B (۵۰mL/μg) and amphotericin B (۴mg/L) were cultured. Then, the plates were incubated at ۳۷°Cunder microaerobic conditions (CO۲ incubator). Urease, catalase and oxidase tests were used for identifyingH. pylori.A bacterial suspension matching the turbidity of a ۲ McFarland standard was poured on the plates. When the surface of the mediumwasdried, sterile paper discs (already autoclaved) were placed on the middle of the mediumwith the help of forceps. ۱۰ microliter of the antibiotic dilution was slowly inoculated onpaper discwith Sampler. The zone of bacteria inhibition wasobserved after ۳ days of incubation in anaerobic condition. Antibacterial effectsof five plants' extracts were tested on resistant bacteria to antibiotics metronidazole.RESULTS AND DISCUSSIONResistance rate to metronidazole was ۶۵%;to ofloxacin, ۲۶%; to ciprofloxacin, ۱۷%;to levofloxacin, ۱۵%;to clarithromycin, ۱۹%; to amoxicillin, ۱۳%; to tetracycline, ۱۲%; and to furazolidone, ۵%. AmongChamaemelum, Glycyrrhizaglabra, Rosmarinus, Hypericumperforatum, Achilleamillefoliunm, Rosmarinus had the most impact on the antibiotic-resistant bacteria.CONCLUSIONAccording to the results of the study and increasing resistance to synthetic antibacterial drug, Rosmarinus plant may be one of the most effective one in removing some bacteria includingH. pylori.
