Investigation of Chitinase۳like-۱ (Chiti۳L۱) Gene Polymorphism (rs۴۹۵۰۹۲۸) with Susceptibility to Allergic Asthma in Iranian Northwestern Azeri Population

Background: In addition to cellular and molecular mechanisms involved in the pathogenesis of asthma, mounting evidences demonstrate that single nucleotide polymorphisms (SNPs) in asthma relevant genes have a role in conferring susceptibility to the disease. CHI۳L۱ is secreted from macrophages, neutrophils, and airway epithelial cells through an IL-۱۳ related mechanism and contributes to tissue remodeling during asthma. Aim of study was to investigate the possible association of rs۴۹۵۰۹۲۸ SNP in Chiti۳L۱ gene with predisposition to allergic asthma in Iranian Northwestern Azeri population. Materials and methods: Frequency of genotypes and alleles of rs۴۹۵۰۹۲۸ SNP in Chiti۳L۱ gene was determined using TaqMan genotyping method in ۱۹۰ patients with asthma and ۱۹۰ healthy controls. Results: Genotype analyzing showed that CC genotype is more frequent (۶۸.۴%) among case group vs ۵۷.۹% in control group while GG genotype is more abundant (۷.۹%) among control group vs ۳.۲% in case group. Furthermore, according to odds ratio (case/ control = ۰.۶۱۱), C allele could be the risk allele whereas G allele can be considered as the protective allele. Conclusion: There is a significant relationship between CHI۳L۱ rs۴۹۵۰۹۲۸ (-۱۳۱ C/G) polymorphism and asthma in studied population (P=۰.۰۳۸, <۰.۰۵). Patients with asthma were mostly found to have C allele whereas most of the healthy individuals had G allele in their genotype.
