Interferon regulatory factor ۷ (IRF۷) gene was cloned from a subtractive cDNA library constructed with mRNAs obtained from rainbow trout (Oncorhynchus mykiss) macrophage cell line (RTS-۱۱). Using expressed sequence tag clones of submitted IRF۷ amino acid sequences, specific primers were designed. Results showed that IRF۷ cDNA contains an ORF of ۱۲۵۱ nucleotides that translates into a ۴۱۶ residues putative peptide. The ۵' untranslated region containing ۱۰۲ nucleotides and the ۳' UTR of the transcript consists of ۶۴۵ nucleotides. The start codon of this ORF is located at nt ۱۰۳ to ۱۰۵ and lies in favorable sequence context for the initiation of translation. Alignment between the rainbow trout IRF۷ and others IRF۷s indicated that the predicted trout IRF۷ association domain was located in residues ۲۰۴-۳۷۱ aa. The rainbow trout IRF۷ nucleotide sequence is most similar to fish IRF۷ with ۴۱% identity for crucian carp, Carssius auratus, ۴۵% for zebrafish, Danio rerio, and to mammalian IRF۷ with ۲۳% for human, Homo sapiens and ۷% for mouse, Mus musculus identity overall.
