Evaluation of the Effect of Serum and Platelet-Rich Plasma (PRP) of Umbilical Cord Blood in Comparison with Extracellular Vesicles (EVs) Derived from them on Skin Rejuvenation In Vitro

چکیده مقاله:

Background and Objective: The process of skin aging in humans is complex and is caused by several factors, including genetics and various environmental factors. In many non-invasive methods of skin rejuvenation, the goal is to restore the elasticity and collagenization of the skin, and for this purpose, various substances are used. The proliferation of fibroblast cells and the production of collagen and other growth factors by these cells is the key to skin rejuvenation. In this research, the effect of umbilical cord blood (and its derivatives) on human fibroblast cells has been investigated. Umbilical cord blood is a biological waste, and in addition to being easy and inexpensive to prepare, it contains a large amount of growth factors and cytokines effective in skin rejuvenation.Materials and Methods: After obtaining umbilical cord blood serum and Platelet-Rich Plasma (PRP), Extracellular Vesicles (EVs) were extracted from them by ultracentrifugation. Then, several tests were performed to confirm and characterize EVs. Next, the effect of these substances and products on cell proliferation and migration and collagen۱ and vimentin protein production in human fibroblast cells was tested.Findings: EVs were confirmed by performing DLS and flow cytometry analysis and BCA kit. Then MTT, scratch assay and immunocytochemistry tests showed that (PRP) ۱۰%, (UBS) ۱۰% and EVs derived from them compared to the control group which contained (FBS)۱۰%, had a better effect on the viability and Cell migration of fibroblast cells and the secretion of collagen۱ and vimentin.Conclusions: The results of this research can be a promise for the use of EVs derived from umbilical cord blood as an off-the-shelf product for skin rejuvenation, which, in addition to being safe and affordable, does not require repeated blood draws from the person.
