Evaluation of miR-۱۸۲, miR-۲۲۱ and CEAExpression in the Peripheral Blood of Oral Squamous CellCarcinoma Patients and Comparing with Healthy Individuals

Background: Head and neck cancer is the sixth common cancerin the world, of which the oral cavity is the most frequenttype. It was diagnosed on more than ۳۷۷,۷۰۰ cases worldwidein ۲۰۲۰. Access to high-quality care leading to a more specificand earlier diagnosis of this cancer is crucial. Therefore,researchers attempt to investigate and detect efficient biologicaltumor marker. The present study aimed to investigate thechanges in the expressions of miR-۱۸۲, miR-۲۲۱, and carcinoembryonicantigen (CEA) in the peripheral blood of patientswith oral squamous cell carcinoma (OSCC) and compare themwith healthy individuals to detect early OSCC.Materials and Methods: ۳۰ peripheral blood samples from patientswith OSCC (۱۹ male and ۱۱ female), aged ۲۵-۷۰, wereobtained from the cancer institute of Tehran University of MedicalSciences, and ۳۰ peripheral blood samples from healthyindividuals (۲۰ male and ۱۰ female) aged ۲۶-۷۰ were collected.Real-time PCR was carried out to investigate the differences inthe expressions of miR-۱۸۲, miR-۲۲۱, and ELISA was used tomeasure CEA protein expression.Results: Among the subjects with OSCC ۸۳% showed miR-۱۸۲ expression, ۹۳% revealed miR-۲۲۱ expression and ۹۶%demonstrated CEA expression. Whereas, these expressionswere ۲۶%, ۲۰% and ۱۳%, respectively, for the healthy group.The simultaneous detection of miR-۱۸۲ and miR-۲۲۱ was ۷۳%in the individuals with OSCC. The simultaneous observationof miR-۱۸۲, miR-۲۲۱, and CEA in the group with OSCC was۶۰%. The expression level of miR-۲۲۱ in the group with OSCCwas ۲.۶۳ times that of the healthy subjects, and the expressionlevel of miR-۱۸۲ in the individuals with OSCC was ۲.۲۹ timesthat of the healthy group.Conclusion: The results of this study nominated miR-۱۸۲,miR-۲۲۱, and CEA as biomarkers for early diagnosis and consequentlyimprovement of survival rate of patients sufferingfrom OSCC.
