Investigating the antiproliferative effectsof oncolytic Coxsackievirus and determining the expressionprofile of microRNA-۲۱ gene in colorectal cancer cells

Background: Oncolytic virotherapy is currently undergoingactive development, which has drawn attention. Research onenteroviruses aims to clarify its pathogenicity. Many therapeutictrials are centered on coxsackievirus A۲۱ (CVA۲۱), whichis a particularly promising candidate for oncolytic virotherapy.MicroRNAs (miRNAs) are a large family of short (۱۹–۲۲ nucleotide)endogenous RNAs that, depending on the target, canact as oncogenes or tumor suppressors. They adversely regulatethe expression of the target genes by cleaving mRNA orby repressing translation. It has been demonstrated that overexpressingmicroRNA-۲۱ significantly lowers the therapeuticeffectiveness of ۵-FU. Determining the expression profile ofthe microRNA-۲۱ gene in colorectal cancer cells and studyingthe antiproliferative effects of oncolytic coxsackievirus are thegoals of the current investigation.Materials and Methods: After HT-۲۹ cell line cultivation, coxsackievirusoncolytic was applied to the cells in multiple MOIs. Next, the cytotoxicity rate, apoptosis rate, and microRNA-۲۱gene expression profile in the HT-۲۹ cell line were assessed.Results: The findings of the current study shown that the oncolyticCoxsackievirus decreases the proliferation rate andmicroRNA-۲۱ expression in HT-۲۹ cells while increasing cellsurvival.Conclusion: The oncolytic Coxsackievirus, which focuses oninterfering with the network of microRNA genes, particularlymicroRNA-۲۱, appears to be a potential anti-colorectal canceragent alongside chemotherapy and radiotherapy, according tothe findings of the current study.
