چکیده مقاله:

Background: Gastric cancer (GC) is one of the malignant tumorswith the highest morbidity and mortality in the world. Earlydiagnosis combined with surgical treatment can significantlyimprove the prognosis of patients. Therefore, it is urgent to seekhigher sensitivity and specificity biomarkers in GC. Long noncodingRNAs (lncRNAs) LncRNAs play a key role in regulatingvarious cellular processes. A number of lncRNAs have beenidentified to be abnormally expressed in gastric cancer. It hasbeen reported that lncRNAs can be regarded as a biomarker andhad particular clinical significance for early screening and GCdiagnosis. Therefore, this study aimed to investigate whetherPLCE۱-AS۲ could be a new diagnostic biomarker in GC.Materials and Methods: In this study, the expression statusof various genes in GC patients was investigated using bioinformaticsanalysis (microarray data analysis downloaded fromthe GEO database). Then, the expression of PLCE۱-AS۲ wasmeasured bioinformatically using the ENCORI database. Aftersample collection, total RNA was extracted using RNA extractionkit in ۲۰ GC tissue samples and ۲۰ adjacent healthy tissuesamples. After cDNA synthesis, real-time PCR technique wasused for evaluating LncRNA PLCE۱-AS۲ expression level inall samples. Results: A real-time PCR experiment evaluated the expressionof potential BC biomarkers found in the bioinformatics analyses.Bioinformatics and experimental analyses revealed thatLncRNA PLCE۱-AS۲ has a remarkable up-regulation in theGC samples. Conclusion: According to obtained results, PLCE۱-AS۲ maybe used as a novel biomarker in gastric cancer.
