Background: With an estimated one million new cases eachyear, gastric cancer (GC) is one of the most frequently diagnosedmalignancies and the third leading cause of cancer-relateddeaths globally. Most individuals with GC are discoveredat advanced stages with a dismal prognosis since they have secretive and unusual symptoms in the early stages. Additionally,circulating lncRNAs can be seen in serum, plasma, andother body fluids when lncRNAs are discharged into the extracellularenvironment. Research interest in circulating lncRNAsand their potential use as biomarkers for cancer diagnosis andprognosis is growing at the moment. Therefore, the goal of thisstudy was to determine whether LINC۰۱۲۸۵ could serve as anovel GC diagnostic biomarker.Materials and Methods: This study examined the status ofmany genes' expression in GC patients using bioinformaticsanalysis (microarray data analysis downloaded from the GEOdatabase). The expression of LINC۰۱۲۸۵ was then computedbioinformatically using the ENCORI database. Using an RNAextraction kit, total RNA was extracted from ۲۰ GC tissuesamples and ۲۰ nearby healthy tissue samples after samplecollection. Following cDNA synthesis, the level of lncRNALINC۰۱۲۸۵ expression in all samples was evaluated using realtimePCR.Results: Real-time PCR was used to assess the expression ofpotential GC biomarkers identified by the bioinformatics investigations.Bioinformatics and experimental studies revealed anamazing dysregulation of the lncRNA LINC۰۱۲۸۵ in the GCsamples.Conclusion: According to the obtained results, lncRNALINC۰۱۲۸۵ can be a suitable diagnostic biomarker for GC.
