The effect of Vesicular stomatitis virusMatrix protein on MMP-۹ gene expression

Breast cancer is the most common cancer that causes deathamong women. Technological advancement in the past threedecades has provided doctors with numerous methods. However,this disease's progression and recurrence rate are stillincreasing. Hence, they are trying to discover more efficientmethods. From the middle of the ۱۹th century, clinical reportsindicated a reduction in tumor recurrence in the case of the virus.Therefore, the possibility of using oncolytic viruses, attenuatedrecombinant viruses, or viruses that have tropism to cancercells is created by genetic engineering methods. Therefore, inthis study, we intend to investigate the effect of VSV oncolyticvirus matrix production, under the survivin promoter, in the expressionof MMP-۹ gene.After the Matrix gene sequence was identified and placeddownstream of Survivin tumor-specific promoter, gene construct(PCDNA۳.۱+M) was synthesized. Using DNA-fectamin,the studied vector was transfected into the MCF-۷ cell line.Thereal-time PCR technique evaluated the effect of the vector containingmatrix gene on the expression of MMP-۹ gene at ۴۸ and۷۲ hours after transfection in the MCF-۷ cell line.Our gene construct can significantly reduce MMP-۹ in ۴۸ and۷۲ hours, which is involved in increased metastasis
