چکیده مقاله:
Background: In the studies, it has been determined that thereare microRNAs in body fluids including sputum. this evidenceled to comprehensive bioinformatics investigations in order tofind microRNAs with high sensitivity and specificity that candetect and differentiate two common subtypes of lung cancer,which are Adenocarcinoma and Squamous cell carcinoma.Based on this, the aim of this research is non-invasive diagnosisof adenocarcinoma and squamous cell carcinoma subgroups oflung cancer based on differential expression of small RNAs.Materials and Methods: The selection of desired microRNAswas done by bioinformatics .For measuring the changes in theexpression of microRNAs, RNA was extracted from the sputumsamples of patients suffering from adenocarcinoma andsquamous cell carcinoma lung cancer. Then the desired cDNAswere synthesized by poly A method. After that, they were subjectedto Real-Time PCR reaction to measure their expressionchanges. To confirm the correctness of the PCR reaction, thePCR reaction products were sequenced. The obtained data werestatistically analyzed by Graphpad prism۸ software.Results: Real-time PCR data showed that the expression levelof miR-۹۴۴ in Squamous cell carcinoma subtype of lung cancercompared to Adenocarcinoma subtype of lung cancer hasincreased significantly (P value: ۰.۰۰۰۱). miR-۳۲۶ has alsoshown a higher level of expression in squamous cell carcinomasubtype of lung cancer than Adenocarcinoma subtype of lungcancer (P value: ۰.۰۴۸۶).Conclusion: This study showed the higher expression level ofmiR-۹۴۴ and miR-۳۲۶ in the sputum samples of squamous cellcarcinoma of the lung compared to the sputum samples of lungadenocarcinoma. From these results, it can be hypothesizedthat with a non-invasive method and the use of the miR۹۴۴ andmiR-۳۲۶ panel, a correct separation of the two common subtypesof lung cancer can be achieved
