P۲۱۹: Ivermectin induces apoptosis in A۵۴۹ lungcancer cells

Objectives: One of the most common causes of death is cancerworldwide and lung cancer is the number one cause of cancerassociateddeath. In most cases, these cancers are uncurable dueto the lack of proper effective medicine, as well as their sideeffects, and we are in need of new kinds of medicines and therapies.Ivermectin can play a wide range of antipathogenic roles.The present study aimed to investigate the effect of ivermectinon the cell viability of A۵۴۹ cancer cells.Materials and Methods: A۵۴۹ cells were cultured in DMEMmedium containing ۱۰% FBS and treated with ۰.۲, ۰.۴, ۰.۸, ۱.۵,and ۸ μM concentration of ivermectin and evaluated by MTT assay for their viability. The expression of Bax, Bad, BCL-۲genes were evaluated by Real-Time PCR. Also, the morphologyof the cells was also examined by Giemsa staining usinginvert microscopy. Data were analyzed using SPSS version ۲۵.Results: Using MTT assay, the IC۵۰ of ivermectin on A۵۴۹cells were determined to be ۰.۸ μM. Highest apoptotic effectwere at ۸ μM killing ۸۶% of cells. In results of Real-Time PCRassay, reduction of BCL-۲ and elevation of Bax and Bad geneswere evident.Conclusion: Ivermectin manifested an excellent apoptotic effecton A۵۴۹ cancer cells. So, it can be concluded that ivermectincan probably be used as an anticancer agent in curing lungcancer.
