Evaluation of miR-۱۴۸, miR-۳۷۵ and Bakgene expression in patients with oral squamous cell carcinomaassociated with oral infection and comparison withOSCC patients without oral infection and healthy individuals

Introduction: MicroRNAs have become recognized as keyplayers in the development of cancer. They are a family of smallnon-coding RNAs that can negatively regulate the expressionof cancer-related genes by sequence-selective targeting of mRNAs,leading to either mRNA degradation or translational repression(۱). Oral squamous cell carcinoma is a multifactorialdisease and the sixth most common cancer in the world (۲).The role of microRNAs in oral cancer has been proven to beinvolved as oncogenes or tumor suppressor genes in this typeof cancer (۳). They are also being studied for their future use as diagnostic and prognostic biomarkers and as potential therapeutictargets (۴).Materials and Methods: In this study, ۳۰ samples of oral cancertissue with infection, ۳۰ samples of oral cancer tissue withoutinfection, and ۶۰ healthy samples were used. Expressionanalysis of miRNA-۱۴۸ and miRNA-۳۷۵ and the BAK genewere performed by Realtime PCR. First, total RNA was extractedand then cDNA was made and the cDNA obtained was usedin Realtime PCR.Results: The expression of microRNA-۱۴۸ was increased incancer samples with infection, compared to cancer sampleswithout infection, and also in cancer samples compared to normalsamples. And the expression of microRNA-۳۷۵ and BAKgene were decreased in cancer samples with infection, comparedto cancer samples without infection, and also in cancersamples compared to normal samples.Conclusion: According to the results, the BAK gene and miRNA-۳۷۵ can help us diagnose early-stage oral cancer by reducingexpression, as well as miRNA-۱۴۸ by increasing expressionas a marker.
