One of the most common diseases that can lead to death is colorectalcancer, which has links to both genetic and environmentalrisk factors. Even though several pathogenic bacteria are linkedto the etiology of colorectal cancer, some others are thought ofas highly selective therapeutic agents for the disease.With multiplesuccessful studies, the use of the bacterial instrument incancer treatment is on its way to becoming a potential methodof colorectal cancer targeted therapy and may one day be a usefulapproach for treating cancer, particularly colorectal cancer.Background: Cancer is the second leading cause of death inthe US, the worldwide cancer epidemic continues to be a publichealth problem. One of the most common life-threateningdiseases is colorectal cancer, which is associated with geneticand environmental risk factors. Even while various pathogenicbacteria are linked to the etiology of colorectal cancer, certainothers are thought of as very selective therapeutic agents for thedisease. Advanced solid tumors might be now be treated withbacterial toxins that have antitumor action as an alternative tochemotherapy.Materials and Methods: In preparation for this review, wesearched databases including PubMed, Google Scholar, and reviewedarticles from ۲۰۱۰ to ۲۰۲۳ Relevant articles about bacterialtoxin therapy and colorectal cancer.Results: Claudin-۳ and Claudin-۴, which are overexpressed incolorectal malignant cell membrane, can directly interact withClostridium perfringens enterotoxin (CPE). A pore will formthrough the membrane as a result of this connection, and theloss of cellular osmotic equilibrium will caused cell death. Recombinantimmunotoxins against cancer have been effectivelyproduced using truncated versions of Diphtheria toxin (DT).An immunotoxin containing gastrin-releasing peptide (GRP,an autocrine growth factor) has been created (DT۳۸۹-GRP) onthe basis of numerous studies demonstrating that the gastrinreleasingpeptide receptor is up regulated on the surface of variousmalignant cell types, including cell lines from small celllung, breast, prostate, and colon cancers. In addition in a mousemodel of sporadic colon cancer caused by inflammation, choleratoxin reduces carcinogenesis.Conclusion: With multiple successful studies, the use of thebacterial instrument in cancer treatment is on its way to becominga potential method of colorectal cancer targeted therapy andmay one day be a useful approach for treating cancer, particularlycolorectal cancer. However further studies needed to optimizeand develop bacterial toxin-based cancer therapy
