Bioinformatics analysis of prognostic valueof CFB as novel key gene in breast cancer

Introduction: Among females, breast cancer (BC) is the mostcommonly diagnosed cancer and the leading cause of cancerdeaths (۱۵%). As breast cancer (BC) is an aggressive malignanttumor with very heterogeneous genomics, exploring genesto serve as biomarkers and therapeutic targets is practical.Complement factor B (CFB) is a component of the alternativepathway of complement activation, and associate with a keysecreted protein to promote proliferation by preventing cellularsenescence. So far, the molecular mechanisms of CFB underlyingthe tumorigenesis and prognosis of BC have not been fully understood yet. Therefore, the findings of the present study mayprovide further understanding of BC development and lead toan improved diagnosis of BC.Materials and Methods: The expression of CFB gene was analyzedby the GEPIA database and UALCAN cancer database.The relationship between CFB expression level and clinicalcharacteristics including prognostic data in breast cancer wereanalyzed by the Kaplan-Meier Plotter and Breast Cancer Gene-Expression Miner (bc-GenExMiner) databases. We also investigatedmutation effects of CFB in breast cancer using TCGAdatabase.Result: We found that CFB gene expression was significantlyupregulated in all histological classification of breast cancer aswell as tumor samples compared to normal tissue. We also havefound that the decreased CFB expression correlated with worserelapse-free survival and overall survival (OS). TCGA mutationanalysis revealed that ۱۰% of patient have loss of function inCFB expression.Conclusion: Therefore, the present study can shed new lighton the understanding of molecular mechanisms of BC and provideCFB as molecular target and diagnostic biomarkers for thetreatment and early diagnosis of breast cancer. More in-depthexperiments and clinical trials are needed to validate the valueof CFB in breast cancer treatment.
