HLA-G is a non-classical HLA class I molecule with immunomodulatory properties that play a dual role in immune system mechanisms by influencing the receptors of innate and adaptive immune cells. Single nucleotide polymorphisms (SNPs) are associated with many disease occurrences. We selected a single nucleotide polymorphism rs۱۰۶۳۳۲۰ including the conversion of an ancestral C to G (MAF=۰.۴۰), which is located at position +۳۱۴۲ of the ۳'UTR of the HLA-G gene. Here, we conducted a case-controlled study to examine the impact of rs۱۰۶۳۳۲۰ in the context of diabetes mellitus type ۲ in the Guilan province. Genomic DNA was extracted from the whole blood samples of all participants. Genotyping was performed using the amplification refractory mutation system PCR (ARMS-PCR) method. The specific primers were designed using Oligo ۷ software. Allele frequencies were determined according to the gene counting method. The strength of the association between the rs۱۰۶۳۳۲۰ poly morphism and diabetes risk was assessed using odds ratios (ORs) and ۹۵% confidence intervals (CIs) for both genotype and allele frequencies. Statistical analyses were performed using SPSS ۲۲.۰. The rs۱۰۶۳۳۲۰ genotype and allele distributions were significantly different in the control and cases groups (P=۰.۰۰۳ and P=۰.۰۰۱, respectively). The GG and GC genotypes serve as the risk factors in the prevalence of diabetes mellitus (OR=۲.۰۵, p=۰.۰۰۲; OR=۱.۹۷, p=۰.۰۰۱, respectively). In conclusion, these results suggested that HLA-G rs۱۰۶۳۳۲۰ may confer individuals’ genetic susceptibility to Diabetes mellitus type ۲. However, further large-scale population-based study is needed to consolidate our findings.
