PHASE SOLUBILITY STUDY OF ATALUREN CYCLODEXTRIN COMPLEXES

Ataluren is a small molecule used for treatment of patients with genetic disorders due to a nonsense mutation. This molecule is an oxadiazole poorly soluble in water, which affect the bioavailability. In the present work, complex formation of this molecule with alpha, beta, gamma, hydroxypropyl and methyl beta cyclodextrin and then the possibility of improving the solubility of Ataluren by this complexation was investigated. The phase solubility studies indicated the formation of Ataluren cyclodextrins complexes at ۱:۱ M ratio in solutions with stability constant of ۵۱۷ M-۱, ۵۸ M-۱, ۱۰۵ M-۱, ۳۵۳ M-۱, and ۴۷۲ M-۱ respectively.
