Using CRISPR-cas۹ technology for inhibition of LncRNAs associated to methamphetamine sensitivity

Major depressive disorder (MDD) is a debilitating and inherited disease in which several lncRNAs are implicated in its etiology. With CRISPR-Cas۱۲, a recently developed system, which selectively disrupts the target site without affecting other DNA regions, we aim to inhibit the expression of three related lncRNAs. We set out to design and use a CRISPR system to disrupt expression of three LncRNA AK۰۳۶۷۹۱, Inc-FANCF-۳ and ACE۱-AS in a mouse model of depression. expression level of AK۰۳۶۷۹۱ was assessed in nucleus accumbens by using quantitative time PCR. Results showed decrease in expression level of all lncRNAs, AK۰۳۶۷۹۱ (p<۰۰۳), Inc-FANCF-۳ (p<۰۰۱) and ACE۱-AS (p<۰۰۱) in nucleus accumbens of test group. It seems that designed CRISPR/Cas۱۲ could induce reduction of expression of genes that are involved in depression. CRISPR/Cas۱۲ also can use for developing knock-in and knock-out mice that may help to shed light to molecular mechanisms of depression in central nervous system.
