Analyzing significant common genes in Parkinson's disease and PI۳K/Akt /mTOR signaling pathway

Introduction and Objective: Parkinson's is a progressive neurodegenerative disorder with symptoms that appear with age. Although numerous researchs have been undertaken to investigate various aspects such as genetics in Parkinson's disease, the disease's primary molecular mechanism remains unexplained. Different signaling pathways are known as factors affecting this disease, among which we can mention dopamine, PI۳K/AKT, and mTOR signaling pathways. PI۳K/AKT signaling pathway plays a very effective role in controlling Parkinson's disease by regulating cell proliferation and apoptosis. On the other hand, the mTOR signaling pathway can prevent the death of nerve cells by regulating energy and creating a balance in autophagy in nerve cells. In this study, we aimed to identify key genes in Parkinson's disease that have a function in PI۳K/AKT/mTOR signaling pathways.Methods: At first, we used the MSigDB database to obtain the gene set of certain signaling pathways. In the second step, we analyzed the GSE۷۶۲۱ dataset. This dataset included ۱۶ patients with Parkinson's disease and ۹ healthy individuals. Then, we shared the genes obtained from both databases. In the end, we drew the gene ontology of the desired genes using the Reactome database.Results: Using the MSigDB database, we found ۱۰۵ genes that play a role in PI۳K/AKT/mTOR pathways. In the next step and after analyzing the GSE۷۶۲۱ dataset, we filtered the genes considering adjusted p value <۰.۰۵ and |Log FC| >۱. After analyzing the data, we observed three key genes that passed all our filters. These genes were VAV۳, CXCR۴ and MAPK۱. Two genes VAV۳ and MAPK۱ showed a significant decrease in expression and CXCR۴ showed a significant increase in expression.Conclusion: In this study, we intended to reveal the common genes of PI۳K/AKT/mTOR signaling pathways in Parkinson's. According to the gene ontology, these genes play an important role in the pathogenesis of Parkinson's disease. Identifying these genes can help in the treatment or diagnosis of this disease.
